THE EFFECT OF PS-K ON THE CARCINOGENESIS OF THE SUBMANDIBULAR GLAND OF RAT


Abstract

The purpose of this study is to investigate the effect of PSK on the carcinogenesis of
salivary gland induced by 9, 10-dimethyl-1,2-benzanthracene(DMBA, Sigma chemical
Co.)
Ninety-six Sprague-Dawley rats, weighing 100-120 gm, were used in this study and
divided into two experimental groups(group 1 and 2) and two control groups(group 3
and 4). 4mg powdered DMBA were injected into the left submandibular gland of sixty
rats(group 1, 2). Thirty rats(group 2) were administered orally 3 times per week with
PSK solution which was dissolved in arachis oil at a dosage of 1mg/200ml. Thirty
rats(group3) were administered with PSK only and group 4, six were untreated.
Five rats from group 1, 2, 3 and one rat from group 4 were sacrificed at 6, 8, 10, 12,
14, 16 weeks after DMBA injection respectively. After sacrificed, the left submandibular
glands were removed, fixed in 10£¥ N-Formalin, embedded in paraffin, sectioned to 4-6
§­ thickness, and stained with Hematoxilin-Eosin stain.
The results were as follows ;
1. DMBA-injected group showed the carcinoma in situ in epithelium which formed the
cyst after 6 to 8 weeks, the infiltrative proliferation after 10weeks, the highly
differentiated squamous cell carcinoma after 12 weeks, and the well differentiated
squamous cell carcinoma after 14 to 16 weeks.
2. 9 cases revealed the fibrosarcoma which was combined with squamous cell carcinoma.
3. Group2 revealed the cystic necrosis and the atrophy of epithelium in all sections, and
also the dilation and the slight proliferative activity of ducts around the cyst after 12 to
16 weeks. It means that group 2 showed the inhibiting effect on carcinogenesis.
4. Group 3 revealed normal features, but showed slight dilation of ducts and congestion
of blood vessels.
